Biological Evaluation of a Native Amniotic Membrane Allograft

This study evaluated the biological properties of a new lyophilized human amnion chorion membrane (LHACM) as a potential wound dressing and surgical tool. The LHACM was compared to the previously used dehydrated amnion/chorion membrane (DHACM) in terms of its biological activity, absorbency, residual hemin levels, and regulatory protein composition.<br /><br />The results showed that LHACM maintained the intrinsic properties and biological activity of amniotic membranes, similar to DHACM. LHACM had improved handling characteristics due to its thicker allograft structure. Histological evaluation confirmed that LHACM retained the natural tissue composition of amniotic membranes.<br /><br />The absorbency of LHACM was measured and compared to DHACM, and the results indicated that LHACM had similar absorbent capacity. Residual hemin levels in LHACM tissue were also measured and found to be comparable to DHACM.<br /><br />The study also assessed the biological activity of LHACM by treating human dermal fibroblasts with LHACM extract. The results showed that LHACM extract promoted cellular proliferation, indicating its potential to support the healing cascade.<br /><br />Furthermore, the LHACM tissue lysates were analyzed for regulatory proteins, and over 350 different factors were identified. These included adhesion receptors, chemokines, ECM molecules, tissue growth factors, inflammatory cytokines, membrane-bound proteins, and signaling receptors.<br /><br />Overall, this study demonstrated that LHACM is a versatile and effective allograft with improved handling characteristics while maintaining the biological properties inherent to amniotic tissues. LHACM has the potential to be widely used in outpatient and surgical applications for wound healing and foot and ankle surgeries.
